jeweils 2 Stdn. Behandlungszeit). - – Hautentfettung: die gewaschene Haut wird dreimal mit reinem Aceton (
jeweils 2 Stdn. Behandlungszeit, Haut-Aceton-Verhältnis 1:5 w/v) unter Rühren extrahiert. - – Aufquellen der Haut: zur nochmaligen Hydratisierung der Haut wird diese nach der Acetonextraktion erneut dreimal mit reinem Wasser (
jeweils 2 Stdn. Behandlungszeit, Haut-Wasser-Verhältnis 1:5 w/v) unter Rühren extrahiert. - – Eiweißabtrennung: nicht kollagene Eiweiße sind aus der Haut abzutrennen. Zu diesem Zweck wird eine Lösung, die 6% NaCl (w/v) und 1% Ameisensäure (w/v) enthält, bereitet. Die aufgequollene Haut wird mit dieser Lösung zweimal unter Rühren extrahiert (

Die Kollagenmenge kann über eine Aminosäureanalyse oder eine Hydroxyprolinbestimmung nach der Methode von Stegemann und Stalder ermittelt werden. Die Bestimmung des Anteils an nativem Kollagen kann mittels CD-Spektroskopie und SDS-PAGE-Elektrophorese erfolgen. Nach Zugabe von 0,1 M Essigsäure bis zu einem Gesamtvolumen von 3 l und Einstellung des pH-Wertes auf 8 bis 9 mittels 10 M NaOH wird das Pepsin gefällt und durch Zentrifugation vom Überstand (Kollagenlösung) abgetrennt. Zur Reinigung der gewonnenen Kollagenlösung wird der pH-Wert auf 3 bis 4 mit Essigsäure eingestellt und das Kollagen mit NaCl (2,5 M NaCl in der Gesamtmischung) ausgefällt und dann abzentrifugiert. Es folgen zwei weitere Reinigungsschritte. In jedem dieser Schritte wird das Kollagen erneut in 0,1 M Essigsäure aufgelöst, mit NaCl (1,8 M NaCl in der Gesamtmischung) wieder ausgefällt und anschließend abzentrifugiert. Um enthaltenes NaCl zu entfernen, wird das gereinigte Kollagen zum Schluss gegen 0,1 M Essigsäure dialysiert (Dialyseschlauch aus regenerierter Cellulose, Porengröße 8–15 kDa).
