2. Polymer nach Anspruch I, dadurch gekennzeichnet, daß Ri eine Cycloalkyldialkylengruppe mit
4 bis 6 annularen Kohlenstoffatomen, wobei die Alkylengruppen 1 bis 4 Kohlenstoffatomen enthalten,
eine C2- bis Qo-Alkylengruppe oder eine C2- bis
Cö-Oxaalkylengruppe mit 1 bis 2 inneren Oxagruppen
bedeuten und R2 und R3 zusammen mit dem
Dioxykohlenstoffatom der Polymerhauptkette einen heteromonocyclischen Ring von 5 bis 6 Gliedern mit
einem Sauerstoffheteroatom, das an das Dioxykohlenstoffatom gebunden ist und die übrigen Glieder
des Ringes Kohlenstoffatome sind, bedeuten.
3. Polymer nach Anspruch 2, dadurch gekennzeichnet, daß R2 und R3 zusammen mit dem
Dioxykohlenstoffatom eine Tetrahydrofurylgruppe bedeuten.
4. Verfahren zur Herstellung der Polymere nach Anspruch 1 bis 3, dadurch gekennzeichnet, daß man
mindestens ei.ie stöchiometrische Menge eines Polyols der allgemeinen Formel:
in der R2 und R3 die oben angegebene Bedeutung
nahen und R6 eine Ci- bis C7-Alkylgruppe ist, in
Gegenwart einer Lewissäure, eines Bronsted- oder eines Organotitanat-Katalysators und gegebenenfalls
in Gegenwart eines inerten Lösungsmittels.
5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, daß man die Reaktion zunächst unter
Atmosphärendruck bei 75 bis ISO0C und anschließend
bei 75 bis 1800C und einem Druck von 0,0001
bis 0,1 mm Hg durchführt.
6. Verwendung der Polymeren nach Anspruch 1 bis 3, zur Herstellung eines biologisch abbaubaren
Formkörpers zur Freisetzung eines Wirkstoffs.
7. Verwendung nach Anspruch 6, wobei der Wirkstoff ein Arzneimittel ist.
Die Umsetzung von Orthoestern mit Glykolen, die zu nichtpolymeren und anderen unterschiedlichen Produkten
führt, ist bekannt aus Veröffentlichungen wie Ind. J. Appl. Chem„ Bd. 28, Nr. 2, S. 53 bis 58,1965, wo Mehrota
et al. Monoäthoxy-monoglykolat und Triglykoxy-bisorthoformiat erhielten durch Umsetzung von Orthoformiat
mit Hexamethylenglykol in molaren Verhältnissen von 1 :1 bis 2 :3. Entsprechend beschreiben Crank et al.
in Aust. J. Chem., Bd. 17, S. 1392 bis 1394, 1964 die Umsetzung von Triolen mit Orthoestern einschließlich
Äthylorthoformiat mit Butan-l,2,4-triol, Pentan-1,2,5-triol
und Pentan-l,3,5-triol unter Bildung monomerer bicyclischer Verbindungen. Bei der Herstellung der
bicyclischen Orthoester durch Umsetzung von Äthylorthoformiat mit Triolen hat es sich gezeigt, daß
Verbindungen, die erhalten worden sind ausgehend von Substanzen mit einer 1,2-Diolstruktur auch Verbindungen
mit Äthylenbindungen ergaben. Entsprechend einer späteren Veröffentlichung (Aust. J. Chem., Bd. 17, S. 1934
bis 1938, 1964) entwickelten Crank et al. diese Umsetzung zu einem synthetischen Verfahren zur
Umwandlung von 1,2-Diolen in Olefine. Später stellte
DeWolf fest, daß Carbonsäure-orthoester reaktionsfähiger sind gegenüber saurer Hydrolyse als die meisten
anderen Verbindungen und diese hohe hydrolytische Reaktionsfähigkeit ihre Synthese und Lagerung erschwert
(Carboxylic Ortho Acid Derivatives, 1970, Academic Press, Inc., New York). Er berichtet, daß die
Umwandlung von Diolen in cyclische Orthoester einschließlich Alkoxydioxolan oder Alkoxvdioxan und
anschließende Hydrolyse ein Verfahren zur Monoacylierung von Diolen darstellt. In letzterer Zeit berichtete
Bailey (Polym. Prepr. Amer. Chem. Soc. Div. Polym.
Chem., Bd. 13, Nr. 1, S. 281 bis 286, 1972), daß die Polymerisation von Spiroorthoestern bei Raumtemperatur
und erhöhten Temperaturen zu Polyestern und Polycarbonaten führte mit den wiederkehrenden
Einheiten

in der R, die oben angegebene Bedeutung hat, umsetzt
mit einer Verbindung der allgemeinen Formel:

Aus Journal of Polymer Science: Polymer Chemistry Edition, Bd. 10,3509-3518 (1972) und Macromolecular
Syntheses', Bd. 4 (1972) Seiten 167 bis 170, sind ebenfalls Polycarbonate bekannt. Diese Polymeren besitzen eine
hohe Stabilität in Wasser und sind nicht -biologisch abbaubar.
Es ist Aufgabe der Erfindung, biologisch abbaubare Polymere zu entwickeln, die als Matrixmaterialien zur
Herstellung biologisch abbaubarer Formkörper geeignet sind.
Diese Aufgabe wird gelöst durch Homo- oder Copolymere, die gekennzeichnet sind durch die
wiederkehrende Einheit der allgemeinen Formel:
-Ri- Ο — C — O-
in der Ri eine C2- bis Cio-Alkylengruppe, eine C2- bis
Cio-AIkepylengruppe, eine C2- bis Ce-Oxaalkylengruppe
mit 1 oder 2 inneren Oxagruppen, eine durch Methylgruppen substituierte Cycloalkylengruppe mit 3
bis 7 annularen Kohlenstoffatomen oder eine cycloalkandialiphatische
Gruppe mit 3 bis 7 annularen Kohlenstoffatomen und 1 bis 7 Kohlenstoffatomen in den Alkylenresten, wobei der Cycloalkanring gegebenenfalls
durch Ci- bis C?-Alkyl- oder C2- bis Cy-Alkenylgruppen
substituiert ist, und R2 und R3 zusammen mit
dem Dioxykohlenstoffatom der Polymerhauptkette eine heteromonocyclische Gruppe mit 5 bis 8 annularen
Gliedern mit 1 oder 2 Sauerstoffheteroatomen, die an das Dioxykohlenstoffatom der polymeren Hauptkette
gebunden sind und deren restliche annulare Glieder Kohlenstoffatome sind, die gegebenenfalls mit Ci- bis
C-7-Alkyl, Cr bis Cr-AIkoxy- oder C2- bis CMlkenylgruppen
substituiert sind, oder wobei an 2 benachbarte Kohlenstoffatome ein Phenylring kondensiert ist, und η
eine ganze Zahl von 10 bis 1000 bedeutet.
Die Erfindung betrifft auch ein Verfahren zur Herstellung der oben beschriebenen Polymere, das
dadurch gekennzeichnet ist, daß man mindestens eine
stöchiometrische Menge eines Polyols der allgemeinen Formel:

in der R1 die oben angegebene Bedeutung hat, umsetzt
mit einer Verbindung der allgemeinen Formel:
R6O-C-OR6
in der R2 und R3 die oben angegebene Bedeutung haben
und R6 eine Ci- bis C7-Alkylgruppe ist, in Gegenwart
einer Lewissäure, eines Brensted- oder eines Organotitanat-Katalysators
und gegebenenfalls in Gegenwart eines inerten Lösungsmittels.
Die Erfindung betrifft auch die Verwendung der oben beschriebenen Polymere als biologisch abbaubare
Matrix für einen Formkörper, der durch den Abbau der Matrix einen Wirkstoff freisetzt.
Die erfindungsgemäßen Polymere umfassen Hornopolymere,
Copolymere mit statistischer Verteilung und Biockcopolymere, die erhalten werden durch Umsetzung
von Monomeren oder Gemischen vorher hergestellter Homopolymere und/oder Copolymere, verzweigte
Polymere und vernetzte Polymere (wenn Ri mehr als zweiwertig ist oder R2 oder R3 eine
vernetzende Gruppe darstellt). Die erfindungsgemäßen Polymere sind thermoplastische lineare Polymere, wenn
Ri zweiwertig ist und R2 und R3 keine vernetzenden
Gruppen darstellt oder aneinander gebunden sind; heiß härtende vernetzte Polymere, wenn Ri zweiwertig ist
und R2 oder R3 eine vernetzende Gruppe ist und heiß härtende vernetzte Polymere, wenn Ri mehr als
zweiwertig ist und R2 und R3 nicht vernetztende
Gruppen darstellen oder miteinander verbunden sind.
Bevorzugte Polymere sind Homo-, Co- und Terpolymere, wobei Rj zweiwertig ist und eine Cycloalkandialkylengruppe
mit 4 bis 6 annularen Kohlenstoffatomen, wobei die Alkylengruppen 1 bis 4 Kohlenstoffatome
enthalten, eine wie oben erwähnte Alkylengruppe oder eine wie oben erwähnte Oxaalkylengruppe bedeuten
und R2 und R3 zusammen mit dem Dioxykohlenstoffatom
der Polymerhauptkette einen heteromonocyclischen Ring mit 5 bis 6 Gliedern mit einem
Sauerstoffheteroatom, das an das Dioxykohlenstoffatom der Polymerhauptkette gebunden ist, bilden,
wobei die restlichen Glieder des Ringes Kohlenstoffa'.ome
bedeuten.
Der Ausdruck »Alkylengruppe«, wie er in dieser Beschreibung für Ri angegeben ist, bedeutet gerad- oder
verzweigtkettige zweiwertige Reste, wie 1,2-Äthylen-,
1,2-Propylen-, 1,7-Heptylen-, 2,3,5-Trimethyl-l,7-heptylen-
und 1,10-Decylengruppen. Der Ausdruck »Alkylengruppe«,
wie er für Rj angewandt wird, bedeutet ungesättigte gerad- oder verzweigtkettige zweiwertige
Reste, wie Vinylen- 2,5-Dimethyl-l,8-oct-3-enylen- und
l,10-Dec-3-enylengruppen. Der Ausdruck »Cycloalkylengruppen«, wie er für Ri angewandt wird, umfaßt
monocyclische Reste, wie Cyclopropylen- und Cycloheptylengruppen.
Beispiele für Cycloalkandialkylen-, Cycloalkandialkenylen-, Cycloalkendialkylen- und Cycloalkendialkenylenreste,
wie sie für Ri angegeben sind, sind Cyclopropandimethylen-, 1,4-Cycloheptandiheptylen-,
Cyclopropandivinylen-, l,5-Cycloheptandihept-4-enylen-, l,3-Cyc!obut-2-en-dimethylen-, 1,3-Cyclohept-5-en-diheptylen-,
LS-Cyclobut^-en-divinylen- und
M-Cyclohept-o-en-dihepM-enylengruppen. Oxaalkylengruppen,
wie sie für Ri angegeben sind, sind beispielsweise Oxaäthylen-, 3-Oxahexylen- und 2,4-Dioxahexylengruppen.
Der Ausdruck »Alkylgruppen«, wie er auftritt bei der Definition gegebenenfalls vorhandener Substituenten
an Resten Ri und heteromonocyclischen Gruppen, wie sie für die Kombination von Rj, R3 und dem
Dioxykohlenstoffatom der Polymerhauptkette angegeben sind, umfaßt gerad- und verzweigtkettige Reste, wie
Methyl-, n-Heptyl-, Isopropyl- und t-Heptylgruppen. Beispiele für Alkenylgruppen, wie sie für die Definition
von R2 und R3 und als gegebenenfalls vorhandener
Substituent an Ri und heteromonocyclischen Gruppen aus einer Kombination von R2, R3 und dem Dioxykohlenstoffatom
der Polymerhauptkette angegeben sind, umfassen die gerad- und verzweigtkettigen Gruppen,
wie Vinyl-, 2-Propenyl-, 1-Heptenyl- und 3-Methyl-4-hexenylgruppen.
Der Ausdruck »Alkoxygruppen«, wie er bei der Definition von R2 und R3 und heteromonocyclisehen
Gruppen aus einer Kombination von R2, R3 und dem Dioxykohlenstoffatom der Polymerhauptkette
angegeben ist, umfaßt die gerad- und verzweigtkettigen Gruppen, wie Methoxy-, n-Heptoxy-, Isopropoxy- und
2-Methylhexoxygruppen. Heteromonocyclische Ringe,
wie sie für die Kombination von R?, R3 und dem
Dioxykohlenstoffatom der Polymerhauptkette angegeben sind, sind beispielsweise die Furylgruppe und ihre
Hydroderivate, Dioxolanyl-, Oxocinyl.gruppen und deren
Hydroderivate und die Dioxocinylgruppe und deren Hydroderivate. Kondensierte hete^opolycyclische Ringe,
wie sie für die Kombination von R2, R3 und dem
Dioxykohlenstoffatom der Polymerhauptkette angegeben sind, sind solche, bei denen ein heterocyi lischer
Ring und ein Phenylring zwei Kohlenstoffatome gemeinem haben, z. B. die Benzfurylgruppe und ihre
Hydroderivate, 4,5-Benz-l,3-dioxolanyl-, 4,5-Benz-l,3-dioxocinylgruppen
und ihre Stellungsisomeren und Hydroderivate und die 4,5-Benzoxacinylgruppe und
deren Stellungsisomere und Hydroderivate.
Alkyigruppen, wie sie für R6 angegeben sind, können
gerad- oder verzweigtkettige Gruppen sein, wie die Methyl-, Isopropyl-, Butyl-, Heptyl- und 3-MethylhexyI-gruppe.
Vorzugsweise ist Rs eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen, d. h. eine Methyl- ois Butylgruppe.
Die einzelnen Gruppen R6 können gleich oder verschieden sein.
Der Ausdruck »wiederkehrende Einheit«, wie er hier verwendet wird, bedeutet die (monomere) Einheit des
Polymers. Zum Beispiel sind die wiederkehrenden Einheiten in einem Homopolymer gleich. Bei einem
Copolymer sind die wiederkehrenden Gruppen unterschiedlich und können in der Polymerkette statistisch
angeordnet sein, wenn die ursprünglichen Einheiten in einem gemeinsamen Reaktionsgefäß copolymerisiert
werden, oder sie können in Blockform angeordnet sein, wenn die Polymere nacheinander gebildet werden,
wobei zunächst eine Homopolymerisation jeder der einzelnen Monomereneinheiten stattfindet. Ein Terpolymer
ist ein Copolymer aus drei verschiedenen wiederkehrenden Einheiten.
Wie oben angegeben, können die erfindungsgemäßen Polymere hergestellt werden durch Umsetzung des
Polyolmonomeren mit dem Ester oder dem Carbonatmonomeren. Die Polymerisationsreaktion wird durchgeführt,
indem man stöchiometrische Mengen der Monomeren oder einen Überschuß des Polyols, d. h.
ungefähr 1 bis 10 Mol Polyol auf 1 Mol Ester oder Carbonatmonomer umsetzt.
Die Polymerisation der Monomeren kann in einem Reaktionsgefäß durchgeführt werden, das mit einem
Rührer und einer Vorrichtung zum Anlegen eines Vakuums versehen ist, unter kontinuierlichem Vermischen
der Monomeren in Gegenwart eines Umesterungskatalysators. Die Polymerisation umfaßt eine
anfängliche Umesterungsreaktion und anschließend eine Polykondensationsreaktion, wobei die gesamte
Polymerisation bei einer Temperatur von 60 bis 220° C und innerhalb von 1 bis 96 h durchgeführt wird. Die
Umesterungsstufe der Reaktion besteht in einem Vermischen der Monomeren mit dem Katalysator unter
kontinuierlichem Rühren des Gemisches und allmählichem Steigern der Temperatur auf 180°C. Die
Umesterungsreaktion der meisten Monomere tritt bei ungefähr 75 bis 180°C innerhalb einer Reaktionszeit von
1 bis 12 h bei normalem Atmosphärendruck unter kontinuierlichem Abdestillieren des Alkohols ein. Die
Polykondensationsreaktion wird begonnen, indem man den Druck auf 0,1 bis 0,001 mm Quecksilber verringert,
wobei die erhöhte Temperatur und der verringerte Druck aufrechterhalten werden und die Reaktionspartner
12 bis 96 h kontinuierlich vermischt werden.
Das Polymer kann unter wasserfreien Bedingungen aus dem Reaktionsgefäß gewonnen werden durch
übliche Isolierungs- und Gewinnungsverfahren. Zum Beispiel kann das Polymer, solange es noch heiß ist,
gewonnen werden durch Extrudieren oder Ausgießen, oder dqs Polymer kann nach dem Abkühlen durch
Lösen in einem trockenen organischen Lösungsmittel, wie Benzo!, Tetrachlorkohlenstoff, Methylenchlorid,
Dioxan, Toluol oder Xylol und anschließende Zugabe einer organischen Flüssigkeit, in der das Polymer
unlöslich ist oder eine begrenzte Löslichkeit hat, zur Ausfällung des Polymers gewonnen werden. Organische
Lösungsmittel für den zuletzt genannten Zweck sind u. a. Äther, Hexan, Pentan, Petroläther und Hexan-Heptan-Gemische.
Das Polymer wird isoliert durch Abfiltrieren und Trocknen unter wasserfreien Bedingungen.
Andere Verfahren zur Gewinnung des Polymers sind z. B. Lyophilisieren aus einem Lösungsmittel.
Typische Umesterungskatalysatoren zur Durchführung der Polymerisationsreaktion sind Lewis-Säuren,
wie Bortrifluorid, Bortrichlorid, Bortrichloridätherat, Bortrifluoridätherat, Zinn-IV-oxychlorid, Phosphoroxychlorid,
Zinkchlorid, Phosphorpentachlorid, Calciumacetat, Antimonmischoxid, Antimonpentachlorid, Antimonpentafluorid,
Zinn-II-ocotoat, Zinn-lV-chlorid, Diäthylzink,
n-ButyllJthium und deren Gemische. Bronsted-Katalysatorpn,
wie p-Toluolsulfonsäure, Polyphosphorsäure,
vernetzte Polystyrolsulfonsäure, saures Silicagel und deren Gemische, Organotitanate, wie Tetrabutyltitanat
und Titannatriumhydrogenhexabutoxid, Oxiden, Carbonate und Acetate von Zink, Calcium, Blei, Magnesium,
Mangan, Kobalt, Alkanoate, Hydride und Alkoxide von Natrium, Lithium, Zink, Calcium, Magnesium und
Aluminium, Antimontrioxid, komplexe Alkoxide und Organomagnesiumhalogenide. Die bevorzugte Menge
des Katalysators beträgt ungefähr 1 Teil Katalysator auf ungefähr 500 Teile des Ester- oder Carbonatmonomeren.
Kleinere oder größere Mengen können ebenfalls angewandt werden, wie 0,005 bis ungefähr 2%, bezogen
auf das Gewicht des Ester- oder Carbonatmonomeren.
Die Polymerisation kann gegebenenfalls in Gegenwart eines inerten organischen Lösungsmittels, das die
Reaktion nicht negativ beeinflußt, durchgeführt werden. Wenn kein zusätzliches Lösungsmittel angewandt wird,
dient einei der Reaktionsteilnehmer, z. B. das Polyol, zunächst als Lösungsmittel. Mit fortschreitender Polymerisation
wird das Lösungsmittelnebenprodukt von dem Reaktionsgemisch durch übliches Abdestillieren,
azeotropes Abdestillieren oder Abdestillieren unter vermindertem Druck entfernt. Geeignete Lösungsmittel
zum azeotropen Destillieren umfassen Toluol, Benzol, m-Xylol, Cumol, Pyridin und n-Heptan.
Die Erfindung wird durch die folgenden Beispiele näher erläutert.
Zu 45 g (0,312 Mol) wasserfreiem trans-1,4-Cyclohexandicarbinol
und 0,05 g Polyphosphorsäure in einem handelsüblichen Polymerisationsreaktor wurden unter
konstantem Rühren unter Stickstoffatmosphäre bei Normaldruck 50 g (0,312 Mol) wasserfreies 2,2 Diäthoxytetrahydrofuran
gegeben. Das Gemisch wurde auf 110 bis 115° C erhitzt und 1,5 bis 2 h unter langsamem
Abdestillieren der entstehenden Flüssigkeit auf dieser Temperatur gehalten. Dann wurde unter Aufrechterhaltung
der Temperatur der Druck nach und nach auf 0,013 mbar verringert und die Temperatur bei diesem
verminderten Druck lanesam auf ISO0C erhöht. Die
Reaktion wurde bei dieser Temperatur 24 h fortgesetzt. Das Polymer wurde isoliert durch Extrudieren aus dem
Reaktionsgefäß. Der Polymer besaß die folgende Struktur, wobei η den Polymerisationsgrad angibt und
10 bis 1000 bedeutet:

und 0,05 g p-Toluolsulfonsäure wurden unter konstantem Rühren 50,5 g (0,312 Mol)
2,2-Dimethoxy-5-methyl-l,3-dioxoIan gegeben und die Polymerisationsreaktion entsprechend Beispiel 1 durchgeführt.

Zu 54,7 g (0,312 Mol) 1,10-Decandiol und 0,05 g
Polyphosphorsäure wurden in einem Reaktionsgefäß unter Stickstoffatmosphäre und Normaldruck unter
konstantem Rühren 50,0 g (0,312 Mol) 2,2-Diäthoxytetrahydrofuran gegeben. Das Gemisch wurde auf 110 bis
115°C erhitzt und 1,5 bis 2 h auf dieser Temperatur gehalten, wobei Äthanol abdestillierte. Dann wurde
unter Aufrechterhaltung der Temperatur der Druck auf 0,013 mbar verringert und bei diesem vermindertem
Druck die Temperatur auf 180° C erhöht. Die Polykondensation wurde 24 h lang durchgeführt Man erhielt ein
Polymer der folgenden Struktur mit einem Polymerisationsgrad von 10 bis 1000:

Die Polykondensation von 2,2-Diäthoxytetrahydrofuran und 30 : 70 eis : trans-Cyclohexandimethanol mit
Polyphosphorsäure wurde folgendermaßen durchgeführt: Das Gewichtsverhältnis des Katalysators zu
Diäthoxytetrahydrofuran betrug 1 :500. Das Monomerverhältnis
von Cyclohexandicarbinol zu Tetrahydrofuran war 2,2 :1, und das Cyclohexandicarbinol wurde als
68%ige Lösung in Methanol in das Reaktionsgefäß gegeben. Das Methanol wurde von dem Diol in situ
abdestilliert und Toluol zugegeben, um Wasser azeotrop von dem System zu entfernen. Das Tetrahydrofuran
wurde dann zugegeben und die Umesterung und Polykondensation bei 18O0C und einem Vakuum von
32 mbar innerhalb von 24 h durchgeführt. Man erhielt eine Ausbeute von 82%. Die GPC-Analyse ergab die
folgenden Werte:

Davon getrennt wurden 14,77 g (0,125 Mol) 1,6-Hexandiol zu 20,0 g (0,125 Mol) 2,2-Diäthoxytetrahydrofuran
und 20 mg p-Toluolsulfonsäure zugegeben

Die erfindungsgemäßen Polymere sind geeignet zur
Herstellung von Vorrichtungen bzw. Abgabekörpern zur Abgabe eines Wirkstoffs an eine wäßrige Umgebung.
Sie besitzen in einer derartigen Umgebung eine steuerbare Hydrophobie, und sie werden kontinuierlich
zu ungiftigen Produkten abgebaut. Der Ausdruck »Abgabe-Körper«, wie er hier gebraucht wird, umfaßt
sowohl Formkörper als auch Produkte, wie Salben, die keine feste Form haben. Die Polymere können nach
bekannten Verfahren zu solchen Abgabekörpern verarbeitet und auf einen Wirkstoff aufgebracht
werden. Zum Beispiel können die Polymere zu Fäden extrudiert, zu Fasern gesponnen, zu Formkörpern
verpreßt, in Lösung zu Folien gegossen und auf einen Wirkstoff durch Verdampfen des Lösungsmittels oder
mit Hilfe eines Wirbelbettes aufgebracht oder gepreßt und/oder geformt oder nach anderen ähnlichen
Standardverfahren verarbeitet werden.
Der Ausdruck »Hydrophobie«, wie er hier und in der weiteren Beschreibung verwendet wird, bezieht sich auf
die Eigenschaft der Polymere, keine nennenswerten Mengen von Wasser zu absorbieren. Die erfindungsgemäßen
hydrophoben Polymere absorbieren Wasser in Mengen, die nicht über 5% ihres Trockengewichtes
hinausgehen.
Die Ausdrücke »abbaubar« und »biologisch abbaubar«, wie sie hier verwendet werden, bezeichnen die
Eigenschaft der Polymeren innerhalb einer Zeitspanne in einer wäßrigen nicht biologischen oder wäßrigen
biologischen Umgebung als Einheit oder insgesamt zu zerfallen. Die Ausdrücke »Abbau« bzw. »Erosion«,
»biologisch abgebaut werden« und »biologischer Abbau« bezeichnen allgemein die Art, nach der der
Abbau bzw. Zerfall des Polymers eintritt
Der Ausdruck »Wirkstoff«, wie er hier in der Beschreibung und den Ansprüchen gebraucht wird,
umfaßt irgendein Mittel, das, wenn es aus dem Abgabekörper freigesetzt wird, zu einem günstigen
Ergebnis führt, z.B. Pesticide, Herbicide, Biocide, Algicide, Rodenticide, Fungicide, Insekticide, Hemmstoffe
für das Pflanzenwachstum, Konservierungsmittel, Kosmetika, Arzneimittel, Mittel zur Verminderung bzw.
Aufhebung oder Begünstigung der Fruchtbarkeit, Luftreiniger und Nährstoffe. Der Ausdruck »Arzneimittel«
umfaßt physiologisch oder pharmakologisch wirksame Substanzen, die zu lokalen oder systemischen
Wirkungen bei Säugetieren führen.
Die Wirkstoffzubereitung innerhalb des Abgabe-Körpers kann in verschiedenen Formen vorliegen, z. B. als
Lösung, Dispersion, Paste, Creme, in Form einzelner Teilchen, Mikrokapseln, Emulsionen und Suspensionen.
Der Abgabekörper kann hergestellt werden, um einen Wirkstoff mit einer Geschwindigkeit nullter
Ordnung oder mit einer variablen Geschwindigkeit freizusetzen, je nach dem Molekulargewicht und der
Zusammensetzung des Polymers, der Konzentration des Wirkstoffs in dem Polymeren und der Oberfläche des
Körpers, die für den Zutritt des umgebenden Mediums freiliegt Vorrichtungen mit unterschiedlicher Freisetzungsgeschwindigkeit
können ebenfalls hergestellt werden durch Verwendung unterschiedlicher Polymere,
die mit verschiedenen Geschwindigkeiten biologisch abgebaut werden oder unter Anwendung verschiedener
Strukturen, wie Laminaten mit unterschiedlichem Wirkstoffgehalt in dem Polymer.
Beispiele für solche Vorrichtungen oder Formkörper zur Freisetzung von Wirkstoffen sind im folgenden
angegeben.
Anwendungsbeispiel 1
Ein plattenförmiger (ellipsoider) Körper (7,5 cm χ 7,5 cm χ 250 μΐη) zur Freisetzung von Hydrocortison
wurde hergestellt durch Vermischen von Hydrocortison mit Poly(2,2-dioxotrans-l,4-cyclohexandimethylentetrahydrofuran)
auf die folgende Weise: 2,375 g Polymer wurden in einem Tiegel auf ungefähr 150° C erhitzt und anschließend 0,125 g feinstteiliges
Hydrocortison zu dem Polymer zugegeben. Es wurde gründlich gemischt, um eine gute Dispersion des
Arzneimittels in dem Polymer zu erhalten. Nach Abkühlen auf Raumtemperatur wurde die Polymer-Arzneimittel-Zubereitung
aus dem Tiegel entnommen und der Körper hergestellt durch 5 Minuten langes Pressen
bei 121°C und 687 daN/cm2 mit Hilfe einer Druckplatte mit einem offenen Bereich von 250 μπι χ 7,5 cm χ 7,5 cm
zwischen Folien aus Polytetrafluorethylen. Der Körper wurde unter einer im wesentlichen trockenen inerten
Atmosphäre mit Hilfe eines Trockenofens hergestellt. Die Oberfläche des Körpers wird in wäßrigem Medium
mit gesteuerter kontinuierlicher Geschwindigkeit von ungefähr 2 μπι Tiefe pro Stunde biologisch abgebaut
Gleichzeitig wird Hydrocortison mit ebenfalls gesteuerter und kontinuierlicher, aber proportionaler Geschwindigkeit
freigesetzt Der Körper kann angewandt werden zur Behandlung von Entzündungen wie Bursitis
(Schleimbeutelentzündung), wenn er auf die Haut oder Schleimhaut aufgebracht wird.
Anwendungsbeispiel 2
Eine Arzneimittel abgebende Salbe wurde folgendermaßen hergestellt:
Zu 2,375 g des viskosen Polymers Poly(2,2-dioxo-l,6-hexamethylentetrahydrofuran)
mit einem Molekulargewicht von ungefähr 25 000, wurden 0,125 g Hydrocortison gegeben und die Bestandteile 5 min gründlich
vermischt Das Vermischen geschah mit Hilfe üblicher Labormischvorrichtungen bei Raumtemperatur und in
trockener inerter Atmosphäre. Die Salbe ist geeignet als lokal anwendbares entzündungshemmendes Mittel.
Anwendungsbeispiel 3
Ein Abgabekörper kann hergestellt werden in Form eines festen Implantats, wie eines Implantats, das ein
Analgetikum freisetzt und angewandt wird bei chirurgischen Operationen, die mit schweren Schmerzen nach
der Operation verbunden sind. In diesen Fällen kann ein Implantat, das ein Analgetikum enthält, in den Körper
eingesetzt werden, während er für die Operation geöffnet ist, um den Schmerz herabzusetzen, da das
Implantat biologisch abgebaut wird und das Analgetikum während der Verheilungszeit freisetzt.
Injizierbare (z. B. durch Trokar-Injektion) Implantate
können ebenfalls hergestellt werden. Ein injizierbares Implantat umfassend ein Polymer mit einer Abbaugeschwindigkeit
von ungefähr 2 μητ/h in einer biologi-
sehen wäßrigen Umgebung mit einem physiologischen pH-Wert von 6 bis 8 und einer Arzneimittelkonzentration
von 5% wurde folgendermaßen hergestellt: Zu 2,375 g Poly(2,2-dioxo-trans-l,4-cyclohexandimethylentetrahydrofuran)
wurden 0,125 g Hydrocortison gegeben und die Bestandteile unter Bildung einer Schmelze
auf 150° C erwärmt. Der Wirkstoff wurde durch 5 Minuten langes Vermischen der Bestandteile in der
Schmelze gut verteilt. Das Mischen wurde unter trockener inerter Atmosphäre unter Normaldruck mit
einer trockenen Vorrichtung vorgenommen. Nachdem das Polymer auf Raumtemperatur abgekühlt war, wurde
es in eine Presse gegeben und durch Spritzgußverfahren zu einem festen zylinderförmigen Implantat mit einem
Durchmesser von 3 mm und einer Länge von 8 mm
geformt. Das Implantat wurde in einen Muskel eines Tieres mit Hilfe eines Trokars eingesetzt, wo es
biologisch abgebaut wurde und das Steroid zur Behandlung der Entzündung freisetzte.
Ein ähnliches Implantat, enthaltend 20% Progesteron in Poly(2,2-dioxo-trans-1 /l-cyclohexandimethylentetrahydrofuran)
mit einem ursprünglichen Gewicht von ungefähr 114 mg besaß in vivo eine Freisetzungsgeschwindigkeit
von Progesteron innerhalb von 7 Tagen von 3.7,3,2,2,9,2,2,2,5 bzw. 3,8 mg/Tag.
Ein anderes injizierbares Implantat enthaltend Norethisteron, wurde hergestellt durch Dispergieren
des Arzneimittels in Poly{2,2-dioxo-trans-1.4-cyclohexandimethylentetrahydrofuran)
nach dem oben angegebenen Verfahren mit der Ausnahme, daß der WirW«tnfr
und das Polymer bei 1300C in trockener Helium-Atmosphäre
vermischt wurden. Das aus diesem Gemisch hergestellte Implantat besaß eine Arzneimittelkonzentration
von 20% und war zylinderförmig. Das Implantat wurde auf beiden Seiten in die paravertebralen Muskeln
von Kaninchen mit Hilfe eines Trokars eingesetzt, wo es mit gesteuerter kontinuierlicher Geschwindigkeit biologisch
abgebaut wurde unter Freisetzung von Norethisteron mit einer Geschwindigkeit von 600 μg/Tag.
Anwendungsbeispiei 4
Es wurden Augeneinsätze hergestellt in Formen wie einer 8 mm Scheibe und einem 6x12 mm Ellipsoid
jeweils mit einer Dicke von 0,4 mm durch Lösen des Arzneimittels und des Polymers in einem trockenen
Lösungsmittel wie Benzol oder 1,4-Dioxan und Lyophilisieren
des Gemisches. Dann wurde das trockene Gemisch bei 100° C unter einem Druck von
1030 daN/cm2 zu einer arzneimittelhaitigen Polymerfolie
verpreßt Nach diesem Verfahren hergestellte Augpneinsätze enthielten 10% Pilocarpinnitrat und
Poly(2^-dioxo-trans-1 ^-cyclohexandimethylentetrahydrofuran),
5% Pilocarpin als freie Base und Poly(2,2-dio-
xo-trans-1 ,^cyclohexajidimeihylentetrahydrofuran),
10% Hydrocortison-Alkohoi und Poly(2£-dioxo-trans-1,4-cyclohexandimethylentetrahydroiiiran),
10% Idoxuridin und Poly(2£-dioxo-trans-i,4-cyclohexandimethylentetrahydrofuran-^-dioxo-oxadimethylen-tetrahydrofuran)
und 10% Chloramphenicol und Poly(2^-di-
oxo-trans-1 ^-cyclohexandimethylentetrahydrofuran-2,2-dioxo-oxadimethylentetrahydrofuran).
Diese Augeneinsätze wurden kontinuierlich biologisch abgebaut und gaben eine abgemessene Menge des
Wirkstoffs an das Auge und das umgebende Gewebe über einen langen Zeitraum ab.
Weitere Augeneinsätze, enthaltend Zinkbacitracin Und das Copolymer mit statistischer Verteilung
Poly(2,2-dioxo-trans-l,4-cyclohexandimethylentetrahydrofuran-2,2-dioxo-l,6-hexamethylentetrahydrofuran)
wurden hergestellt durch 5 Minuten langes Vermischen des Bacitracins bei 120° C in der heißen Schmelze des
Polymers. Nach Abkühlen auf Raumtemperatur wurde die Arzneimittel-Polymer-Zubereitung bei 1200C unter
einem Druck von 687 daN/cm2 innerhalb von 5 Minuten zu einer Folie verpreßt. Aus dieser Folie wurden
Augeneinsätze hergestellt. Die Freisetzungsgeschwindigkeit von Augeneinsätzen innerhalb von 3 Tagen, die
aus der oben angegebenen Arzneimittel-Polymer-Zubereitung hergestellt worden waren, ist in der folgenden
Tabelle angegeben:

Anwendungsbeispiel 5
Es können auch Auflagen mit dem wirkstoffhaltigen Polymer hergestellt werden, wie solche, die für
Verbrennungen und zur Wundbehandlung angewandt werden, indem die Masse (Wirkstoff-Polymer) in e;nen
absorbierenden Träger eingebaut wird (es können Träger, wie Stoffe, Plastik, Fasern, Schaumstoffe oder
Papier angewandt werden). Typische Wirkstoffe, die in solchen Auflagen verwendet werden können, sind die
verschiedenen Sulfa-Arzneimittel, Salicylsäuredenvate und andere Antibiotika Zum Beispiel werden in einen
runden Ausschnitt aus einem doppelt gewirkten Polyesterstoff 4 g Poly(2,2-dioxohexamethylentetrahydrofuran),
enthaltend 0.1 bis 30, üblicherweise 2 bis 15% «-Amino-p-toluolsulfoniimid eingebracht, indem man
das ivirkstorrhaltige Polymer mit einem Spatel in den
Stoff einstreicht Ahnlich kann das hydrophobe Polymer
mit anderen Wirkstoffen zubereitet werden, wie 1 bis 1,5% Silbernitrat in Poly(2^-dioxohexamethylentetrahydrofuran),
0,001 bis 2 Gew.-% Collagenase mit 1 bis 1,5 Gew.-% Silbernitrat in Poly(2^-dioxohexamethylentetrahydrofuran)
und 1 g Poly(2£-dioxohexamethylentetrahydrofuran),
enthaltend 82XlO3 Caseineinheiten
einer Protease, wie Streptokinase oder ähnliches.
