Abstract
Description
The invention relates to an incubator, in particular for Polymerase chain method, using at least two Storage containers, each one specific to one Contains temperature adjustable liquid and with a Holding device for incubation cuvettes, the successively optionally with the liquids of the Storage containers can be brought into contact.
The polymerase chain method is one newly developed method of molecular biology with which it is possible any base sequence of a DNA Section of a genetic information that is either from a virus, a bacterium or any cells higher living beings can originate, in many simple ways to duplicate. Through this multiple duplication of one Base sequence are gain factors of over one hundred thousand achievable. This is the base sequence in a quantity that is conventional with chemical Methods can be demonstrated in a simple manner.
The polymerase chain method usually requires that successive incubation of a reagent batch at least three different temperatures, each Incubation step performed for a period of time which can take a few seconds to a few minutes. The different temperatures of the incubation steps differ significantly in some cases, with a correspondingly rapid temperature changes must take place. E.g. there is a first incubation step during a Time between a few seconds to a few minutes 95 ° C, followed by a second incubation step at z. B. 20 ° C, which is also a few seconds to a few minutes lasts. This incubation step is followed, for example, by a third Incubation step at a temperature between 55 ° C and can be 70 ° C. This third incubation step too can take a few seconds to a few minutes. Doing so usually the duration of the individual incubation steps precisely defined. Usually the succession these three at different temperatures Incubation steps taking place with appropriate predetermined incubation times repeated many times. In the Such incubation steps are, for example. repeated thirty times.
In a known incubator for the polymerase chain Method come at least two, preferably three Storage containers for use arranged side by side are, and each of which is a specific one Contains temperature adjustable liquid. This known incubator has one for the incubation cuvettes provided holding device, which is designed as a basket in which the incubation cuvettes are attached. A Robot, which is controlled by a computer, is used to the holding device designed as a basket for the Incubation cuvettes from one storage container to the next Transport storage container and the holding device with the incubation cuvettes in the respective Liquid located during a storage tank immerse in a certain period of time. The time spans during which the incubation cuvettes in the on a liquid at a certain temperature are set using the computer. These known method requires in addition to the storage containers and the holding device for the incubation cuvettes Robots or a variety of individual devices and is out because of this, relatively expensive. Another shortcoming exists in that this well-known incubator in its application is comparatively cumbersome.
There is also damage to the incubation cuvettes during its conversion from a storage container to one not reliably exclude neighboring storage containers, if this implementation should be carried out in a time-saving manner.
The present invention is based on the object to create an incubator of the type mentioned, the is simple, with damage to the Incubation cuvettes can be safely avoided, and at what temperature changes on the order of up to at 100 ° C can be achieved within a few seconds.
This object is achieved in that an incubation chamber for receiving the holding device for the incubation cuvettes that everyone Storage container by means of a pipeline with the Incubation chamber is connected that each pipe has a Has valve device, and that a device for optional transport of the liquid between the Incubation chamber and the storage containers is provided.
The incubation cuvettes are made using the Holding device arranged in the incubation chamber, see above that an implementation of the incubation cuvettes no longer is required to set up the incubation cuvettes with a liquid in a certain temperature To get in touch. This will damage the Incubation cuvettes avoided. The application of Incubation cuvettes at a certain temperature located liquid takes place in that the corresponding storage containers by means of the associated Pipeline fluidly connected to the incubation chamber is so that the located in this reservoir and a certain temperature liquid from Storage container can flow to the incubation chamber. Around only a certain storage container with the To fluidly connect the incubation chamber, everyone has Pipeline a valve device. The facility for optional transport of the liquid between the Incubation chamber and one of the storage containers provided the appropriate liquid within from one of the storage containers to To direct the incubation chamber to the in the Incubation cuvettes with the liquid at a certain temperature wash around and thus the incubation cuvettes inside bring to the appropriate temperature in the shortest possible time. After performing the appropriate incubation step this liquid is transformed into a liquid by the device Liquid transport back out of the incubation chamber diverted. Immediately afterwards the liquid can another storage container with a suitable one Switch position of the valve device of the individual Pipelines to carry out the next Incubation step initiated in the incubation chamber will. This procedure can be varied to repeat.
Each storage container preferably has one Flow line and a return line for connection with the incubation chamber in which the Valve devices are provided.
It has proven to be advantageous that all Flow lines with a first common manifold and all return lines with a second common manifold are connected, the first and the second manifold with the incubation chamber are connected, and the facility for Liquid transport in the first and / or in the second Bus line is arranged. With the help of the first common manifold are all Flow lines interconnected in a star configuration. Accordingly, using the second joint Collector line all return lines star-shaped connected with each other. Through the two manifolds the connection between the one incubation chamber and the number of storage containers for those on certain, different from each other Liquids. Through the It is also training with the two manifolds easily possible, a single facility for Liquid transport between the incubation chamber and to be provided in the various storage containers. It is if necessary, the facility for Liquid transport either in the first or in the to provide second manifold. Such training can be sufficient if the between each Storage containers and the common incubation chamber not transporting amounts of liquid per unit of time too large, or if the cross section of the two Manifolds is large enough. To be particularly advantageous However, it has been found that the facility for Liquid transport is one pump, the two Has pump elements, the one pump element in the first manifold and the second pump element in the second manifold is arranged. In this way it is possible with a common drive unit to drive both pump elements together, whereby per Unit of time, as desired, relatively large amounts of liquid between the incubation chamber and the individual Storage containers can be pumped around.
The provided in the individual pipes Valve devices are preferably as solenoid valves educated. Solenoid valves of this type have the advantage that they are simply controlled by electrical means, d. H. can be operated as desired. Another The advantage of such solenoid valves is that they have good switching behavior so that it the individual piping is possible as required shut off or open within a short period of time in order to as desired between the incubation chamber and one certain reservoir a fluid connection to manufacture.
Each storage container preferably has a volume that is larger than the volume of the incubation chamber. On this way temperature shifts are caused by the Heat capacity of the incubation chamber and through the Heat capacity of those in the incubation chamber Incubation cuvettes or the holding device for the Incubation cuvettes avoided, so that exact Temperature jumps are possible. This results exactly reproducible parameter values for the exact execution of the Polymerase chain method.
The incubator according to the invention has at least one Storage container on a heater. Preferably each individual storage container with a heating device educated. This heater can be trade electric heating elements.
It has proven to be expedient to have at least one To form storage containers with a cooling device. Of course, it is also possible to have a larger one Number of storage containers or all storage containers with one Training cooling device. With such a Cooling device, it is possible in the corresponding Liquid in the reservoir as required a certain temperature below the ambient temperature cool down. In practical use with a Such cooling device temperatures down to -30 ° C can be generated be.
To generate ultrasonic waves inside the Incubation chamber can be attached to the incubation chamber Ultrasound generator can be arranged. With such a Ultrasonic generator is possible in the Incubation chamber or in the incubation chamber incubation cuvettes or reagent tubes a molecular movement with the reagents contained therein generate which is the molecular biological reaction can accelerate considerably. It can do the same purpose serve to generate microwave radiation inside the incubation chamber to the incubation chamber Microwave generator, especially a magnetron to be arranged. This makes it possible to microwave in the incubation chamber radiate, on the one hand, to rapid heating of the reagents in the incubation cuvettes can contribute, on the other hand, by not thermal effects of microwave radiation Molecular movement in the incubation cuvettes located reagents can be generated.
Each incubator reservoir can hold one Have temperature sensors. With these temperature sensors is a monitoring of the liquid filling of the individual Storage container, d. H. monitoring the temperature of the liquid in the individual storage containers possible. With the help of the appropriate Temperature sensor determined temperature value from the given Setpoint, either the associated Heating device or the associated cooling device of the corresponding reservoir activated.
Each storage container can be equipped with a level sensor be trained. With these level sensors it is possible to monitor the filling quantity of each storage container and to regulate if necessary.
It has proven to be advantageous that a tax and Control device is provided with the Valve devices, the pump, the heating devices, the Cooling devices, the ultrasonic generator, the Microwave generator, the temperature sensors or the Level sensors is connected. Preferably the Control and regulating device via a microprocessor controlled. The microprocessor can have a RAM for permanent storage of a large number of Incubation programs, as well as an input keyboard for the Microprocessor to call the corresponding Incubation process parameters. A special software for the microprocessor in conjunction with the Input keyboard and a display for easy entry all desired process data such as temperature levels, Sequence of the storage containers to be selected and number the temperature-time cycles to be run. The Process parameters are used for a variety of possible incubation programs permanently in the RAM of the Microprocessor stored. This RAM can be trade a so-called battery-buffered RAM.
The microprocessor preferably assigns a fixed program automatic query of the process parameters. The Different programs can be selected by pressing any key be called. Advantageously, therefore Input of the process parameters no programming knowledge required because that is in the microprocessor Fixed program the query of the necessary parameters in the Dialogue between the display and the user of the incubator one after the other. The connection of the tax and Control device with the microprocessor allowed also exact process control in terms of temperature and time compliance individual incubation steps. At process termination or at It is the occurrence of a malfunction of the incubator possible that with the help of an alarm device, the is also connected to the microprocessor acoustic and / or optical alarm signal is given.

With the aid of the ultrasound generator 56 , it is possible to generate a microwave radiation inside the incubation chamber 12 , with the aid of which the reagents in the incubation cuvettes 52 are excited to increase their temperature-related molecular movement. The microwave generator 58 , which is provided on the incubation chamber 12 and serves, for example, a magnetron known per se, serves the same purpose. With the help of the microwaves generated by the microwave generator 58 , the reagents located in the incubation cuvettes 52 are also excited to accelerate their molecular movement.

The connection between the ultrasound generator 56 provided on the incubation chamber 12 and the regulating and control device 60 is indicated by the connecting line 76 . An electrical connecting line 78 is arranged between the microwave generator 58 provided on the incubation chamber 12 and the control and regulating device 60 .
With the aid of the control and regulating device 60 having a microprocessor, it is possible to regulate the temperature of the liquid 16 in the individual storage containers 14 as desired. For this purpose, the actual value of the temperature of the liquid 16 in the corresponding storage container 14 is determined with the aid of the corresponding temperature sensor 14 and, if necessary, the temperature of the liquid 16 to a predetermined desired value with the aid of the heating device 46 or cooling device 48 controlled by the control and regulating device 60 regulated. With the aid of the fill level sensors 42 , it is possible to determine the fill level of the liquid 16 of each storage container 14 and to compare the actual value determined in this way in the control and regulating device 60 with a predetermined target value and to use the control and regulating device 60 to control the magnetic valves 22 in this way switch that the actual value of the level in each storage container 14 is brought into agreement with a predetermined target value. Of course, the drive unit 38 is actuated simultaneously with the aid of the control and regulating device 60 , as a result of which the two pump elements 26 and 32 come into operation. With the help of the control and regulating device 60 , it is thus possible to let the liquids 16 of the individual storage containers 14 flow back and forth between the storage containers 14 and the common incubation chamber 12 within a very short time, in order to sequentially carry the reagents located in the incubation cuvettes 52 to apply the liquids 16 , which are at different temperatures. This application can take place within precisely defined time periods, whereby these time periods can also be determined by means of the control and regulating device 60 having a microprocessor. To further accelerate the molecular movement of the reagents located in the incubation cuvettes 52, the ultrasound generator 56 and / or the microwave generator 68 can also be activated with the aid of the control and regulating device 60 .
