- Die vorliegende Erfindung betrifft die Verabreichung von Arzneimitteln gegen erektile Dysfunktion über den Inhalationsweg. Spezifisch betrifft sie Aerosole, die Arzneimittel gegen erektile Dysfunktion enthalten und in der Inhalationstherapie eingesetzt werden.
- Hintergrund der Erfindung
- Es gibt eine Reihe von Zusammensetzung, die derzeit zur Behandlung von erektiler Dysfunktion auf dem Markt sind. Die Zusammensetzungen enthalten zumindest einen Wirkstoff, der die beobachteten therapeutischen Wirkungen bereitstellt. Zu den Wirkstoffen in solchen Zusammensetzungen gegen erektile Dysfunktion gehören Sildenafil, Tadalafil und Vardenafil.
- Es ist wünschenswert, einen neuen Verabreichungsweg für Arzneimittel gegen erektile Dysfunktion bereitzustellen, das rasch hohe Konzentrationen der Verbindungen im Plasma bereitstellt. Die Bereitstellung eines solchen Weges ist ein Ziel der vorliegenden Erfindung.
- Zusammenfassung der Erfindung
- Die vorliegende Erfindung betrifft die Verabreichung von Arzneimitteln gegen erektile Dysfunktion auf dem Inhalationsweg. Spezifisch betrifft sie Aerosole, die Arzneimittel gegen erektile Dysfunktion enthalten und in der Inhalationstherapie eingesetzt werden.
- In einem Zusammensetzungsaspekt der vorliegenden Erfindung umfasst das Aerosol zumindest 5 Gew.-% eines Arzneimittels gegen erektile Dysfunktion. Vorzugsweise umfassen die Teilchen zumindest 10 Gew.-% eines Arzneimittels gegen erektile Dysfunktion. Noch bevorzugter umfassen die Teilchen zumindest 20 Gew.-%, 30 Gew.-%, 40 Gew.-%, 50 Gew.-%, 60 Gew.-%, 70 Gew.-%, 80 Gew.-%, 90 Gew.-%, 95 Gew.-%, 97 Gew.-%, 99 Gew.-%, 99,5 Gew.-% oder 99,97 Gew.-% eines Arzneimittels gegen erektile Dysfunktion.
- Typischerweise handelt es sich bei dem Arzneimittel gegen erektile Dysfunktion nicht um Sildenafilcitrat.
- Typischerweise weist das Aerosol eine Masse von zumindest 10 μg auf. Vorzugsweise weist das Aerosol eine Masse von zumindest 100 μg auf. Noch bevorzugter weist das Aerosol eine Masse von zumindest 200 μg auf.
- Typischerweise umfassen die Teilchen weniger als 10 Gew.-% Abbauprodukte des Arzneimittels gegen erektile Dysfunktion. Vorzugsweise umfassen die Teilchen weniger als 5 Gew.-% Abbauprodukte des Arzneimittels gegen erektile Dysfunktion. Noch bevorzugter umfassen die Teilchen weniger als 2,5, 1, 0,5, 0,1 oder 0,03 Gew.-% Abbauprodukte des Arzneimittels gegen erektile Dysfunktion.
- Typischerweise umfassen die Teilchen weniger als 90 Gew.-% Wasser. Vorzugsweise umfassen die Teilchen weniger als 80 Gew.-% Wasser. Noch bevorzugter umfassen die Teilchen weniger als 70 Gew.-%, 60 Gew.-%, 50 Gew.-%, 40 Gew.-%, 30 Gew.-%, 20 Gew.-%, 10 Gew.-% oder 5 Gew.-% Wasser.
- Typischerweise sind zumindest 50 Gew.-% des Aerosols in amorpher Form vorhanden, worin kristalline Formen weniger als 50 Gew.-% des gesamten Aerosol-Gewichts ausmachen, unabhängig von der Natur der einzelnen Teilchen. Vorzugsweise sind zumindest 75 Gew.-% des Aerosols in amorpher Form vorhanden. Noch bevorzugter sind zumindest 90 Gew.-% des Aerosols in amorpher Form vorhanden.
- Typischerweise weist das Aerosol eine Dichte der inhalierbaren Aerosolteilchen von mehr als 106 Teilchen/ml auf. Vorzugsweise weist das Aerosol eine Dichte der inhalierbaren Aerosolteilchen von mehr als 107 Teilchen/ml oder 108 Teilchen/ml auf.
- Typischerweise weisen die Aerosolteilchen einen massenmittleren aerodynamischen Durchmesser von weniger als 5 μm auf. Vorzugsweise weisen die Teilchen einen massenmittleren aerodynamischen Durchmesser von weniger als 3 μm auf. Noch bevorzugter weisen die Teilchen einen massenmittleren aerodynamischen Durchmesser von weniger als 2 oder 1 μm auf.
- Typischerweise beträgt die geometrische Standardabweichung um den massenmittleren aerodynamischen Durchmesser der Aerosolteilchen weniger als 3,5. Vorzugsweise beträgt die geometrische Standardabweichung weniger als 3,0. Noch bevorzugter beträgt die geometrische Standardabweichung weniger als 2,5 oder 2,2.
- Typischerweise wird das Aerosol durch das Erhitzen einer Zusammensetzung, die ein Arzneimittel gegen erektile Dysfunktion enthält, um Dampf zu bilden, und nachfolgend durch die Kondensation des Dampfes zu einem Aerosol gebildet.
- In einem anderen Zusammensetzungsaspekt der vorliegenden Erfindung umfasst das Aerosol Teilchen, die zumindest 5 Gew.-% Sildenafil, Tadalafil oder Vardenafil umfassen. Vorzugsweise umfassen die Teilchen zumindest 10 Gew.-% Sildenafil, Tadalafil oder Vardenafil. Noch bevorzugter umfassen die Teilchen zumindest 20 Gew.-%, 30 Gew.-%, 40 Gew.-%, 50 Gew.-%, 60 Gew.-%, 70 Gew.-%, 80 Gew.-%, 90 Gew.-%, 95 Gew.-%, 97 Gew.-%, 99 Gew.-%, 99,5 Gew.-% oder 99,97 Gew.-% Sildenafil, Tadalafil oder Vardenafil.
- Typischerweise weist das Aerosol eine Masse von zumindest 10 μg auf. Vorzugsweise weist das Aerosol eine Masse von zumindest 100 μg auf. Noch bevorzugter weist das Aerosol eine Masse von zumindest 200 μg auf.

- Wenn das Aerosol Sildenafil umfasst, weist es typischerweise eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 5 mg/l und 40 mg/l auf. Vorzugsweise weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 10 mg/l und 35 mg/l auf. Noch bevorzugter weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 15 mg/l und 30 mg/l auf.
- Wenn das Aerosol Tadalafil umfasst, weist es typischerweise eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 2,5 mg/l und 20 mg/l auf. Vorzugsweise weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 3,5 mg/l und 17,5 mg/l auf. Noch bevorzugter weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 5 mg/l und 15 mg/l auf.
- Wenn das Aerosol Vardenafil umfasst, weist es typischerweise eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 1 mg/l und 20 mg/l auf. Vorzugsweise weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 1,5 mg/l und 17,5 mg/l auf. Noch bevorzugter weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 2 mg/l und 15 mg/l auf.
- Typischerweise weist das Aerosol eine Dichte der inhalierbaren Aerosolteilchen von mehr als 106 Teilchen/ml auf. Vorzugsweise weist das Aerosol eine Dichte der inhalierbaren Aerosolteilchen von mehr als 107 Teilchen/ml oder 108 Teilchen/ml auf.
- Typischerweise weisen die Aerosolteilchen einen massenmittleren aerodynamischen Durchmesser von weniger als 5 μm auf. Vorzugsweise beträgt die geometrische Standardabweichung weniger als 3,0. Noch bevorzugter beträgt die geometrische Standardabweichung weniger als 2,5 oder 2,2.
- Typischerweise wird das Aerosol durch das Erhitzen einer Zusammensetzung, die Sildenafil, Tadalafil oder Vardenafil enthält, um Dampf zu bilden, und die folgende Kondensation des Dampfes zu einem Aerosol gebildet.
- In einem Verfahrensaspekt der vorliegenden Erfindung wird ein Arzneimittel gegen erektile Dysfunktion einem Säugetier über den Inhalationsweg verabreicht. Das Verfahren umfasst Folgendes: a) Erhitzen einer Zusammensetzung, wobei die Zusammensetzung zumindest 5 Gew.-% eines Arzneimittels gegen erektile Dysfunktion umfasst, zur Bildung eines Dampfs; und b) Ermöglichung des Abkühlens des Dampfes, wobei ein Kondensationsaerosol gebildet wird, das Teilchen umfasst und von dem Säugetier inhaliert wird. Vorzugsweise umfasst die Zusammensetzung, die erhitzt wird, zumindest 10 Gew.-% des Arzneimittels gegen erektile Dysfunktion. Noch bevorzugter umfasst die Zusammensetzung zumindest 20 Gew.-%, 30 Gew.-%, 40 Gew.-%, 50 Gew.-%, 60 Gew.-%, 70 Gew.-%, 80 Gew.-%, 90 Gew.-%, 95 Gew.-%, 97 Gew.-%, 99 Gew.-%, 99,5 Gew.-%, 99,9 Gew.-% oder 99,97 Gew.-% eines Arzneimittels gegen erektile Dysfunktion.
- Typischerweise umfassen die Teilchen zumindest 5 Gew.-% eines Arzneimittels gegen erektile Dysfunktion. - Typischerweise weist das Aerosol eine Dichte der inhalierbaren Aerosolteilchen von mehr als 106 Teilchen/ml auf. Vorzugsweise weist das Aerosol eine Dichte der inhalierbaren Aerosolteilchen von mehr als 10 Teilchen/ml oder 108 Teilchen/ml auf.
- Typischerweise beträgt die Rate der Bildung von inhalierbaren Aerosolteilchen aus dem zugeführten Kondensationsaerosol mehr als 108 Teilchen/s. Vorzugsweise wird das Aerosol in einer Rate von mehr als 109 inhalierbaren Teilchen/s gebildet. Noch bevorzugter wird das Aerosol in einer Rate von mehr als 1010 inhalierbaren Teilchen/s gebildet.
- Typischerweise wird das zugeführte Kondensationsaerosol in einer Rate von mehr als 0,5 mg/s gebildet. Vorzugsweise wird das Aerosol in einer Rate von mehr als 0,75 mg/s gebildet. Noch bevorzugter wird das Aerosol in einer Rate von mehr als 1 mg/s, 1,5 mg/s oder 2 mg/s gebildet.
- Typischerweise führt die Zufuhr des Kondensationsaerosols in weniger als 1 h zu einer Spitzenplasmakonzentration eines Arzneimittels gegen erektile Dysfunktion in dem Säugetier. Vorzugsweise wird die Spitzenplasmakonzentration in weniger als 0,5 h erreicht. Noch bevorzugter wird die Spitzenplasmakonzentration in weniger als 0,2, 0,1, 0,05, 0,02, 0,01 oder 0,005 h (arterielle Messung) erreicht.
- In einem weiteren Verfahrensaspekt der vorliegenden Erfindung werden Sildenafil, Tadalafil oder Vardenafil einem Säugetier über den Inhalationsweg verabreicht. Das Verfahren umfasst Folgendes: a) Erhitzen einer Zusammensetzung, wobei die Zusammensetzung zumindest 5 Gew.-% Sildenafil, Tadalafil oder Vardenafil umfasst, zur Bildung eines Dampfs; und b) Ermöglichung des Abkühlens des Dampfes, wobei ein Kondensationsaerosol gebildet wird, das Teilchen umfasst und von dem Säugetier inhaliert wird. Vorzugsweise umfasst die Zusammensetzung, die erhitzt wird, zumindest 10 Gew.-% Sildenafil, Tadalafil oder Vardenafil. Noch bevorzugter umfasst die Zusammensetzung zumindest 20 Gew.-%, 30 Gew.-%, 40 Gew.-%, 50 Gew.-%, 60 Gew.-%, 70 Gew.-%, 80 Gew.-%, 90 Gew.-%, 95 Gew.-%, 97 Gew.-%, 99 Gew.-%, 99,5 Gew.-%, 99,9 Gew.-% oder 99,97 Gew.-% Sildenafil, Tadalafil oder Vardenafil.

- Typischerweise umfassen die Teilchen weniger als 10 Gew.-% Abbauprodukte von Sildenafil, Tadalafil oder Vardenafil. - Typischerweise sind zumindest 50 Gew.-% des Aerosols in amorpher Form vorhanden, worin kristalline Formen weniger als 50 Gew.-% des gesamten Aerosol-Gewichts ausmachen, unabhängig von der Natur der einzelnen Teilchen. - Typischerweise weisen die Aerosolteilchen einen massenmittleren aerodynamischen Durchmesser von weniger als 5 μm auf. Vorzugsweise weisen die Teilchen einen massenmittleren aerodynamischen Durchmesser von weniger als 3 μm auf. Noch bevorzugter weisen die Teilchen einen massenmittleren aerodynamischen Durchmesser von weniger als 2 oder 1 μm auf.
- Typischerweise beträgt die geometrische Standardabweichung um den massenmittleren aerodynamischen Durchmesser der Aerosolteilchen weniger als 3,5. Vorzugsweise beträgt die geometrische Standardabweichung weniger als 3,0. Noch bevorzugter beträgt die geometrische Standardabweichung weniger als 2,5 oder 2,2.
- Wenn das Aerosol Sildenafil umfasst, weist das verabreichte Aerosol typischerweise eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 5 mg/l und 40 mg/l auf. Vorzugsweise weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 10 mg/l und 35 mg/l auf. Noch bevorzugter weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 15 mg/l und 30 mg/l auf.
- Wenn das Aerosol Tadalafil umfasst, weist das verabreichte Aerosol typischerweise eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 2,5 mg/l und 20 mg/l auf. Vorzugsweise weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 3,5 mg/l und 17,5 mg/l auf. Noch bevorzugter weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 5 mg/l und 15 mg/l auf.
- Wenn das Aerosol Vardenafil umfasst, weist das verabreichte Aerosol typischerweise eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 1 mg/l und 20 mg/l auf. Vorzugsweise weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 1,5 mg/l und 17,5 mg/l auf. Noch bevorzugter weist das Aerosol eine Massendichte des inhalierbaren Aerosolarzneimittels zwischen 2 mg/l und 15 mg/l auf.
- Typischerweise weist das Aerosol eine Dichte der inhalierbaren Aerosolteilchen von mehr als 106 Teilchen/ml auf. Vorzugsweise weist das Aerosol eine Dichte der inhalierbaren Aerosolteilchen von 107 Teilchen/ml oder 108 Teilchen/ml auf.
- Typischerweise beträgt die Rate der Bildung von inhalierbaren Aerosolteilchen aus dem zugeführten Kondensationsaerosol mehr als 108 Teilchen/s. Vorzugsweise wird das Aerosol in einer Rate von mehr als 109 inhalierbaren Teilchen/s gebildet. Noch bevorzugter wird das Aerosol in einer Rate von mehr als 1010 inhalierbaren Teilchen/s gebildet.
- Typischerweise wird das zugeführte Kondensationsaerosol in einer Rate von mehr als 0,5 mg/s gebildet. Vorzugsweise wird das Aerosol in einer Rate von mehr als 0,75 mg/s gebildet. Noch bevorzugter wird das Aerosol in einer Rate von mehr als 1 mg/s, 1,5 mg/s oder 2 mg/s gebildet.
- Wenn das Kondensationsaerosol Sildenafil umfasst, werden dem Säugetier bei einer Einatmung typischerweise zwischen 5 mg und 40 mg Sildenafil zugeführt. Vorzugsweise werden dem Säugetier bei einer Einatmung zwischen 10 mg und 35 mg Sildenafil zugeführt. Noch bevorzugter werden bei einer Einatmung zwischen 15 mg und 30 mg Sildenafil zugeführt.
- Wenn das Kondensationsaerosol Tadalafil umfasst, werden dem Säugetier bei einer Einatmung typischerweise zwischen 2,5 mg und 20 mg Tadalafil zugeführt. Vorzugsweise werden dem Säugetier bei einer Einatmung zwischen 3,5 mg und 17 mg Tadalafil zugeführt. Noch bevorzugter werden bei einer Einatmung zwischen 5 mg und 15 mg Tadalafil zugeführt.
- Wenn das Kondensationsaerosol Vardenafil umfasst, werden dem Säugetier bei einer Einatmung typischerweise zwischen 1 mg und 20 mg Vardenafil zugeführt. Vorzugsweise werden dem Säugetier bei einer Einatmung zwischen 1,5 mg und 17,5 mg Vardenafil zugeführt. Noch bevorzugter werden bei einer Einatmung zwischen 2 mg und 15 mg Vardenafil zugeführt.
- Typischerweise führt die Zufuhr des Kondensationsaerosols in weniger als 1 h zu einer Spitzenplasmakonzentration von Sildenafil, Tadalafil oder Vardenafil in dem Säugetier. Vorzugsweise wird die Spitzenplasmakonzentration in weniger als 0,5 h erreicht. Noch bevorzugter wird die Spitzenplasmakonzentration in weniger als 0,2, 0,1, 0,05, 0,02, 0,01 oder 0,005 h (arterielle Messung) erreicht.
- In einem Set-Aspekt der vorliegenden Erfindung wird ein Set zur Verabreichung eines Arzneimittels gegen erektile Dysfunktion an ein Säugetier über den Inhalationsweg bereitgestellt, wobei das Set Folgendes umfasst: a) eine Zusammensetzung, die zumindest 5 Gew.-% eines Arzneimittels gegen erektile Dysfunktion umfasst; und b) eine Vorrichtung, die aus dieser Zusammensetzung ein Aerosol des Arzneimittels gegen erektile Dysfunktion bildet, zur Inhalation durch ein Säugetier. Vorzugsweise umfasst die Zusammensetzung zumindest 20 Gew.-%, 30 Gew.-%, 40 Gew.-%, 50 Gew.-%, 60 Gew.-%, 70 Gew.-%, 80 Gew.-%, 90 Gew.-%, 95 Gew.-%, 97 Gew.-%, 99 Gew.-%, 99,5 Gew.-%, 99,9 Gew.-% oder 99,97 Gew.-% des Arzneimittels gegen erektile Dysfunktion.
- Typischerweise umfasst die Vorrichtung, die Teil des Sets ist: a) ein Element zum Erwärmen des Arzneimittels gegen erektile Dysfunktion zur Bildung eines Dampfes; b) ein Element, welches das Abkühlen des Dampfes zur Bildung eines Aerosols ermöglicht; und c) ein Element, das die Inhalation des Aerosols durch das Säugetier ermöglicht.

- Der "aerodynamische Durchmesser" eines bestimmten Teilchens bezieht sich auf den Durchmesser eines kugelförmigen Tröpfchens mit einer Dichte von 1 g/ml (Dichte von Wasser), das dieselbe Absetzgeschwindigkeit wie das Teilchen aufweist.
- "Aerosol" bezieht sich auf eine Suspension von festen oder flüssigen Teilchen in einem Gas.
- Die "Massendichte des Aerosolarzneimittels" bezieht sich auf die Sildenafil- oder Tadalafilmasse pro Volumeneinheit des Aerosols.

- Ein geeignetes Verfahren wird eingesetzt, um die Aerosole der vorliegenden Erfindung zu bilden. Ein bevorzugtes Verfahren umfasst das Erhitzen einer Zusammensetzung, die ein Arzneimittel gegen erektile Dysfunktion enthält, zur Bildung eines Dampfes, gefolgt von dem Abkühlen des Dampfes, so dass er kondensiert und ein Aerosol (Kondensationsaerosol) bereitgestellt wird, das ein Arzneimittel gegen erektile Dysfunktion umfasst. Die Zusammensetzung wird in einer von vier Formen erhitzt: als reine aktive Verbindung (z.B. reines Sildenafil, Tadalafil oder Vardenafil); als Gemisch einer aktiven Verbindung und eines pharmazeutisch annehmbaren Arzneimittelträgers; als Salzform einer reinen aktiven Verbindung; und als Gemisch eines Salzes einer aktiven Verbindung und eines pharmazeutisch annehmbaren Arzneimittelträgers.
- Salzformen von Arzneimitteln gegen erektile Dysfunktion (z.B. Sildenafil, Tadalafil oder Vardenafil) sind entweder im Handel erhältlich oder werden aus der entsprechenden freien Base unter Einsatz von Verfahren, die auf dem Gebiet der Erfindung bekannt sind, erhalten. Verschiedene pharmazeutisch annehmbare Salze sind für die Aerosolierung geeignet. Solche Salze umfassen, ohne Einschränkung, Folgende: Salze der Salzsäure, Bromwasserstoffsäure, Essigsäure, Maleinsäure, Ameisensäure und Fumarsäure.
- Pharmazeutisch annehmbare Arzneimittelträger können flüchtig oder nicht-flüchtig sein. Flüchtige Arzneimittelträger werden bei Erhitzen gleichzeitig verflüchtigt, aerosoliert und mit dem Arzneimittel gegen erektile Dysfunktion inhaliert. Solche Arzneimittelträgerklassen sind auf dem Gebiet der Erfindung bekannt und umfassen, ohne Einschränkung, gasförmige, überkritisch flüssige, flüssige und feste Lösungsmittel. Es folgt eine Liste beispielhafter Träger dieser Klassen: Wasser; Terpene, wie z.B. Menthol; Alkohole, wie z.B. Ethanol, Propylenglykol, Glycerin und weitere ähnliche Alkohole; Dimethylformamid; Dimethylacetamid; Wachs; überkritisches Kohlendioxid; Trockeneis und Gemische davon.
- Feste Träger, auf denen die Zusammensetzungen erhitzt werden, haben verschiedene Formen. Beispiele umfassen, ohne Einschränkung, Zylinder mit weniger als 1,0 mm Durchmesser, Behälter mit weniger als 1,0 mm Dicke und fast jede Form, die von kleinen (z.B. weniger als 1,0 mm großen) Poren durchdrungen ist. Vorzugsweise stellen feste Träger ein hohes Verhältnis von Oberfläche zu Volumen (z.B. mehr als 100 pro Meter) und ein hohes Verhältnis von Oberfläche zu Masse (z.B. mehr als 1 cm2 pro Gramm) bereit.
- Ein fester Träger einer Form kann auch in eine andere Form mit anderen Eigenschaften umgewandelt werden. Eine flache, 0,25 mm dicke Bahn weist beispielsweise ein Verhältnis von Oberfläche zu Volumen von etwa 8.000 pro Meter auf. Durch das Aufrollen der Bahn zu einem hohlen Zylinder mit 1 cm Durchmesser entsteht ein Träger, der weiterhin das hohe Verhältnis von Oberfläche zu Masse der ursprünglichen Bahn aufweist, aber ein geringeres Verhältnis von Oberfläche zu Volumen aufweist (etwa 400 pro Meter).
- Eine Reihe von verschiedenen Materialien werden eingesetzt, um die festen Träger zu erzeugen. Solche Materialien gehören, ohne Einschränkung, zu den folgenden Klassen: Metalle, anorganische Materialien, kohlenstoffhältige Materialien und Polymere. Es folgen Beispiele für diese Materialklassen: Aluminium, Silber, Gold, Edelstahl, Kupfer und Wolfram; Siliciumdioxid, Glas, Silicon und Aluminiumoxid; Graphit, poröse Kohlenstoffe, Kohlenstoffgarne und Kohlenstofffilze; Polytetrafluorethylen und Polyethylenglykol. Kombinationen von Materialien und beschichteten Varianten der Materialien werden ebenfalls eingesetzt.
- Wenn Aluminium als fester Träger eingesetzt wird, ist Aluminiumfolie ein geeignetes Material. Beispiele für Materialien auf Siliciumdioxid-, Aluminiumoxid- und Siliconbasis umfassen amorphes Siliciumdioxid S-5631 (Sigma, St. Louis, Mo.), BCR171 (ein Aluminiumoxid mit einer definierten Oberfläche von mehr als 2 m2/g von Aldrich, St. Louis, Mo.) und einen Siliconwafer, wie in der Halbleiterindustrie eingesetzt. Kohlenstoffgarne und -filze können von American Kynol, Inc., New York, NY, bezogen werden. Chromatographieharze, wie z.B. chemisch an poröses Siliciumdioxid gebundenes Octadecylsilan, sind beispielhafte beschichtete Varianten von Siliciumdioxid.
- Das Erhitzen der Zusammensetzungen des Arzneimittels gegen erektile Dysfunktion erfolgt durch ein beliebiges geeignetes Verfahren. Beispiele für Verfahren, durch die Hitze erzeugt werden kann, umfassen Folgende: Leiten eines Stroms durch ein elektrisches Widerstandselement; Absorption von elektromagnetischer Strahlung, wie z.B. Mikrowellen oder Laserlicht; sowie exotherme chemische Reaktionen, wie z.B. exotherme Solvatation, Hydratation von pyrophoren Materialien und Oxidieren von brennbaren Materialien.
- Zufuhr von Aerosolen, die Arzneimittel gegen erektile Dysfunktion enthalten.
- Aerosole der vorliegenden Erfindung, die Arzneimittel gegen erektile Dysfunktion enthalten, werden einem Säugetier unter Einsatz einer Inhalationsvorrichtung zugeführt. Wenn das Aerosol ein Kondensationsaerosol ist, weist die Vorrichtung zumindest drei Elemente auf: ein Element zum Erhitzen einer Zusammensetzung, die ein Arzneimittel gegen erektile Dysfunktion umfasst, zur Bildung eines Dampfes; ein Element, das das Abkühlen des Dampfes ermöglicht, wodurch ein Kondensationsaerosol bereitgestellt wird; und ein Element, das die Inhalation des Aerosols durch ein Säugetier ermöglicht. Verschiedene geeignete Verfahren zum Erhitzen sind obenstehend beschrieben. Das Element, das das Abkühlen ermöglicht, ist in seiner ein fachsten Form ein inerter Durchgang, der das Heizmittel mit dem Inhalationsmittel verbindet. Das Element, das die Inhalation ermöglicht, ist eine Aerosolausgangsöffnung, die eine Verbindung zwischen dem Abkühlelement und dem Atemsystem des Säugetiers bildet.
- Eine Vorrichtung, die zur Zufuhr des Aerosols, das das Arzneimittel gegen erektile Dysfunktion umfasst, eingesetzt wird, wird unter Bezugnahme auf
1 beschrieben. Die Zufuhrvorrichtung100 weist ein proximales Ende102 und ein distales Ende104 , ein Heizelement106 , eine Stromquelle108 und ein Mundstück110 auf. Eine Zusammensetzung, die ein Arzneimittel gegen erektile Dysfunktion enthält, wird auf einer Oberfläche112 des Heizelements106 aufgebracht. Bei Aktivierung eines durch den Benutzer betätigten Schalters114 initiiert die Stromquelle108 das Erhitzen des Heizelements106 (z.B. durch Entzündung eines brennbaren Brennstoffs oder das Leiten von Strom durch ein Widerstandsheizelement). Die Arzneimittelzusammensetzung gegen erektile Dysfunktion verflüchtigt sich aufgrund des Erhitzens des Heizelements106 und kondensiert zur Bildung eines Kondensationsaerosols, bevor sie das Mundstück110 am proximalen Ende der Vorrichtung102 erreicht. Ein Luftstrom, der sich von dem distalen Ende der Vorrichtung104 zu dem Mundstück110 bewegt, trägt das Kondensationsaerosol zum Mundstück110 , wo es von dem Säugetier inhaliert wird. - Nach Wunsch umfassen die Vorrichtungen verschiedene Komponenten, um die Zufuhr des Aerosols, das ein Arzneimittel gegen erektile Dysfunktion enthält, zu erleichtern. Die Vorrichtung kann beispielsweise eine beliebige Komponente umfassen, die auf dem Gebiet der Erfindung zur Steuerung des Zeitpunkts der Arzneimittelaerosolierung bezogen auf die Inhalation (z.B. Betätigung durch Atmung), zur Bereitstellung von Feedback an Patienten in Bezug auf die Rate und/oder das Volumen der Inhalation, zur Vermeidung übermäßiger Verwendung (z.B. "Sperr"-Element), zur Vermeidung des Einsatzes durch nicht autorisierte Individuen und/oder zur Aufzeichnung der Dosierungsgeschichte bekannt sind.
- Dosierung von Aerosolen, die Sildenafil oder Tadalafil enthalten.
- Die Dosierungsmenge eines Arzneimittels gegen erektile Dysfunktion in Aerosolform entspricht im Allgemeinen nicht mehr als der doppelten Standarddosis bei oraler Verabreichung des Arzneimittels. Sildenafil, Tadalafil und Vardenafil werden beispielsweise zur Behandlung von erektiler Dysfunktion in einer Stärke von 25 mg, 10 mg bzw. 5 mg verabreicht. In Aerosolform werden im Allgemeinen für dieselbe Indikation 5 bis 40 mg Sildenafil, 2,5 bis 20 mg Tadalafil und 1 bis 20 mg Vardenafil verabreicht. Eine typische Dosierung eines Aerosols eines Arzneimittels gegen erektile Dysfunktion wird in einer einzigen Inhalation oder als eine Reihe von Inhalationen innerhalb einer Stunde oder weniger (Dosierung entspricht der Summe der inhalierten Mengen) verabreicht. Wenn das Arzneimittel in einer Reihe von Inhalationen verabreicht wird, kann eine unterschiedliche Menge bei jeder Inhalation verabreicht werden.
- Die geeignete Dosis eines Aerosols, das ein Arzneimittel gegen erektile Dysfunktion enthält, zur Behandlung einer bestimmten Erkrankung kann unter Einsatz von Verfahren, wie z.B. Tierversuchen und klinischen Versuchen zur Dosisbestimmung (Phase I/II), ermittelt werden. Ein Tierversuch umfasst das Messen der Plasmakonzentrationen des Arzneimittels in einem Tier, nachdem diesem das Aerosol verabreicht wurde. Säugetiere, wie z.B. Hunde oder Primaten, werden typischerweise für solche Untersuchungen eingesetzt, da ihr Atmungssystem dem menschlichen Atmungssystem ähnlich ist. Die Anfangsdosis für Versuche bei Menschen ist im Allgemeinen niedriger als die Dosis in dem Tiermodell, die zu Plasmaarzneimittelspiegeln in Zusammenhang mit einer therapeutischen Wirkung bei Menschen führte, oder entspricht dieser. Dann wird eine Dosisanpassung bei Menschen durchgeführt, bis eine optimale therapeutische Reaktion erhalten wird oder eine Toxizität auftritt, die die Dosis begrenzt.
- Analyse von Aerosolen, die Arzneimittel gegen erektile Dysfunktion enthalten.
- Die Reinheit eines Aerosols, das ein Arzneimittel gegen erektile Dysfunktion enthält, wird unter Einsatz von verschiedenen Verfahren bestimmt, wobei Beispiele für diese Verfahren von Sekine et al., Journal of Forensic Science 32, 1271-1280 (1987), und Martin et al., Journal of Analytic Toxicology 13, 158-162 (1989), beschrieben werden. Ein Verfahren umfasst die Bildung eines Aerosols in einer Vorrichtung, wobei ein durch diese verlaufender Gasstrom (z.B. Luftstrom), im Allgemeinen in einer Rate von 0,4 bis 60 l/min, aufrechterhalten wird. Der Gasstrom trägt das Aerosol in einer oder mehrere Auffangvorrichtungen. Nach der Isolation aus der Auffangvorrichtung wird das Aerosol einem Analyseverfahren, wie z.B. einer Gas- oder Flüssigchromatographie, unterzogen, das die Bestimmung der Reinheit der Zusammensetzung ermöglicht.
- Verschiedene Auffangvorrichtungen werden zum Auffangen des Aerosols eingesetzt. Die folgende Liste umfasst Beispiele für solche Auffangvorrichtungen: Filter; Glaswolle; Impinger; Lösungsmittelauffangvorrichtungen, wie z.B. Trockeneis-gekühlte Ethanol-, Methanol-, Aceton- und Dichlormethanauffangvorrichtungen, Niederdruckbehälter (z.B. Vakuumbehälter), in die das Aerosol gezogen wird; sowie leere Behälter, die die Aerosol-Erzeugungsvorrichtung vollständig umgeben und abschließen. Wenn ein Feststoff, wie z.B. Glaswolle, eingesetzt wird, wird dieser typischerweise mit einem Lösungsmittel, wie z.B. Ethanol, extrahiert. Das Lösungsmittelextrakt wird dann eher als der Feststoff (z.B. Glaswolle) selbst einer Analyse unterzogen. Wenn eine Spritze oder ein Behälter eingesetzt werden, wird der Behälter auf ähnliche Weise mit einem Lösungsmittel extrahiert.
- Die oben erläuterte Gas- oder Flüssigchromatographie umfasst ein Nachweissystem (d.h. einen Detektor). Solche Nachweissysteme sind auf dem Gebiet der Erfindung bekannt und umfassen beispielsweise Flammenionisations-, Photonenabsorptions- und Massenspektrometrie-Detektoren. Ein Vorteil einer Massenspektrometrie-Detektors besteht darin, dass sie zur Bestimmung der Struktur der Abbauprodukte des Arzneimittels gegen erektile Dysfunktion eingesetzt werden kann.
- Die Teilchengrößenverteilung eines Aerosols, das ein Arzneimittel gegen erektile Dysfunktion enthält, wird unter Einsatz eines beliebigen, auf dem Gebiet der Erfindung bekannten Verfahrens ermittelt (z.B. Kaskadenimpaktion). Ein achtstufigen Non-Viable-Kaskadenimpaktor von Andersen (Andersen Instruments, Smyrna, Ga.), der durch eine Scheinöffnung (USP-Öffnung, Andersen Instruments, Smyrna, GA) mit einem Ofenrohr verbunden ist, ist ein System, das für Kaskadenimpaktionsuntersuchungen herangezogen wird.
- Die Massendichte des inhalierbaren Aerosols wird beispielsweise durch die Zufuhr eines Arzneimittel-hältigen Aerosols in eine abgeschlossene Kammer über eine Inhalationsvorrichtung und das Messen der in der Kammer gesammelten Masse bestimmt. Typischerweise wird das Aerosol durch das Vorhandensein eines Druckgradienten zwischen der Vorrichtung und der Kammer in die Kammer gezogen, wobei die Kammer unter geringerem Druck steht als die Vorrichtung. Das Volumen der Kammer sollte etwa dem Atemzugsvolumen eines inhalierenden Patienten entsprechen.
- Die Massendichte eines inhalierbaren Aerosol-Arzneimittels wird beispielsweise durch die Zufuhr eines Arzneimittel-hältigen Aerosols in eine abgeschlossene Kammer über eine Inhalationsvorrichtung und das Messen der in der Kammer gesammelten Menge der aktiven Arzneimittelverbindung bestimmt. Typischerweise wird das Aerosol durch das Vorhandensein eines Druckgradienten zwischen der Vorrichtung und der Kammer in die Kammer gezogen, wobei die Kammer unter geringerem Druck steht als die Vorrichtung. Das Volumen der Kammer sollte etwa dem Atemzugsvolumen eines inhalierenden Patienten entsprechen. Die Menge der in der Kammer gesammelten aktiven Arzneimittelverbindung wird durch das Extrahieren der Kammer, das Durchführen einer chromatographischen Analyse des Extrakts und den Vergleich der Ergebnisse der chromatographischen Analyse mit jenen eines Standards, der bekannte Mengen des Arzneimittels enthält, bestimmt.
- Die Teilchendichte des inhalierbaren Aerosols wird beispielsweise durch die Zufuhr eines Aerosolphasen-Arzneimittels in eine abgeschlossene Kammer über eine Inhalationsvorrichtung und das Messen der Anzahl der in der Kammer gesammelten Teilchen mit einer bestimmten Größe bestimmt. Die Anzahl der Teilchen mit einer bestimmten Größe kann unmittelbar basierend auf den lichtstreuenden Eigenschaften der Teilchen gemessen werden. Alternativ dazu wird die Anzahl der Teilchen mit einer bestimmten Größe durch das Messen der Masse der Teilchen in einem bestimmten Größenbereich und die Berechnung der Anzahl der Teilchen basierend auf der Masse wie folgt ermittelt: Gesamtanzahl der Teilchen = Summe (von Größenbereich 1 bis Größenbereich N) der Anzahl der Teilchen in jedem Größenbereich. Die Anzahl der Teilchen in einem bestimmten Größenbereich = Masse des Größenbereichs/Masse eines typischen Teilchens in diesem Größenbereich. Masse eines typischen Teilchens in einem bestimmten Größenbereich = π·D3·φ/6, wobei D ein typischer Teilchendurchmesser in dem Größenbereich (im Allgemeinen der Mittelwert der Grenz-MMAD, die den Größenbereich bestimmen) in μm, φ die Teilchendichte (in g/ml) ist und die Masse in Pikogramm-Einheiten (g–12) angegeben wird.
- Die Rate der Bildung inhalierbarer Aerosolteilchen wird beispielsweise durch die Zufuhr eines Aerosolphasen-Arzneimittels in eine abgeschlossene Kammer über eine Inhalationsvorrichtung bestimmt. Die Zufuhr erfolgt über einen bestimmten Zeitraum (z.B. 3 s) hinweg, und die Anzahl der Teilchen mit einer bestimmten Größe, die in der Kammer gesammelt wurden, wird wie oben erläutert bestimmt. Die Rate der Teilchenbildung entspricht der Anzahl der gesammelten, 100 nm bis 5 μm großen Teilchen dividiert durch die Dauer der Sammelzeit.
- Die Rate der Aerosolbildung wird beispielsweise durch die Zufuhr eines Aerosolphasen-Arzneimittels in eine abgeschlossene Kammer über eine Inhalationsvorrichtung bestimmt. Die Zufuhr erfolgt über einen bestimmten Zeitraum (z.B. 3 s) hinweg, und die Masse an gesammeltem Teilchenmaterial wird durch das Wiegen der abgeschlossenen Kammer vor und nach der Zufuhr des Teilchenmaterials bestimmt. Die Rate der Aerosolbildung entspricht dem Anstieg der Masse in der Kammer dividiert durch die Dauer der Sammelzeit. Alternativ dazu kann, wenn eine Veränderung der Masse der Zufuhrvorrichtung oder einer Komponente davon nur durch die Freisetzung von Aerosolphasen-Teilchenmaterial verursacht werden kann, die Masse des Teilchenmaterials mit der verlorenen Masse der Vorrichtung oder einer Komponente davon während der Zufuhr des Aerosols gleichgesetzt werden. In diesem Fall entspricht die Rate der Aerosolbildung der Abnahme der Masse der Vorrichtung oder einer Komponente davon während der Zufuhr dividiert durch die Dauer der Zufuhr.
- Die Rate der Arzneimittelaerosolbildung wird beispielsweise durch die Zufuhr eines Aerosols, das ein Arzneimittel gegen erektile Dysfunktion enthält, in eine abgeschlossene Kammer über eine Inhalationsvorrichtung über einen bestimmten Zeitraum (z.B. 3 s) hinweg bestimmt. Wenn es sich bei dem Aerosol um ein reines Arzneimittel gegen erektile Dysfunktion handelt, wird die Menge des in der Kammer gesammelten Arzneimittels wie oben beschrieben bestimmt. Die Rate der Arzneimittelaerosolbildung entspricht der Menge des in der Kammer gesammelten Arzneimittels gegen erektile Dysfunktion dividiert durch die Dauer der Sammelzeit. Wenn das Aerosol, das das Arzneimittel gegen erektile Dysfunktion enthält, einen pharmazeutisch annehmbaren Arzneimittelträger enthält, stellt das Multiplizieren der Aerosolbildungsrate mit dem Prozentsatz des Arzneimittels gegen erektile Dysfunktion in dem Aerosol die Rate der Arzneimittelaerosolbildung bereit.
- Nutzen von Aerosolen, die Arzneimittel gegen erektile Dysfunktion enthalten.
- Aerosole der vorliegenden Erfindung, die Arzneimittel gegen erektile Dysfunktion enthalten, werden typischerweise zur Behandlung von erektiler Dysfunktion eingesetzt.
- Die folgenden Beispiele dienen der Veranschaulichung, und nicht der Einschränkung, der vorliegenden Erfindung.
- Sildenafilcitrat ist im Handel als Wirkstoff in VIAGRA® erhältlich und kann unter Einsatz von auf dem Gebiet der Erfindung bekannten Standardverfahren isoliert werden.
- Tadalafil kann unter Einsatz der in
US-Patent Nr. 6.143.746 (veröffentlicht am 7. November 2000), das hiermit durch Verweis hierin aufgenommen wird, beschriebenen Verfahren synthetisiert werden. Vardenafil kann unter Einsatz der inWO 99/24433 - Beispiel 1
- Allgemeines Verfahren zum Erhalt der freien Base einer Salzverbindung
- Etwa 1 g Salz (z.B. Monohydrochlorid) wird in entionisiertem Wasser (~30 ml) gelöst. Drei Äquivalente Natriumhydroxid (1N NaOHaq) werden zu der Lösung zugetropft, und der pH wird überprüft, um sicherzustellen, dass diese basisch ist. Die wässrige Lösung wird 4-mal mit Dichlormethan (~50 ml) extrahiert, und die Extrakte werden kombiniert, getrocknet (Na2SO4) und filtriert. Die filtrierte organische Lösung wird unter Einsatz eines Rotationsverdampfers eingeengt, um die gewünschte freie Base bereitzustellen. Bei Bedarf erfolgt die Reinigung der freien Base unter Einsatz von Standardverfahren, wie z. B. Chromatographie oder Umkristallisieren.
- Beispiel 2
- Allgemeines Verfahren zur Verflüchtigung von Verbindungen aus einer Halogenlampe
- Eine Lösung des Arzneimittels in etwa 120 μl Dichlormethan wird auf ein 3,5 cm × 7,5 cm großes Stück Aluminiumfolie (zuvor mit Aceton gereinigt) beschichtet. Das Dichlormethan wird verdampfen gelassen. Die beschichtete Folie wird um eine 300-Watt-Halogenröhre (Feit Electric Company, Pico Rivera, Calif.) gewunden, die in eine an einem Ende mit einem Gummistöpsel verschlossene Glasröhre eingeführt wird. Das 2,5 s lange Leiten von 90 V Wechselstrom (durch eine durch einen Steiltransformator gesteuerte Leitung) durch die Lampe stellt thermischen Dampf (umfassend Aerosol) bereit, der an den Glasröhrenwänden gesammelt wird. Umkehrphasen-HPLC-Analyse mit Nachweis durch Absorption von 225-nm-Licht wird eingesetzt, um die Reinheit des Aerosols zu bestimmen. (Nach Wunsch wird das System vor der Verflüchtigung mit Argon gespült.)
- Tadalafilaerosol (0,29 mg) wurde unter Einsatz dieses Verfahrens mit einer Reinheit von 98,5 % erhalten. Um reinere Aerosole zu erhalten, kann eine geringere Menge des Arzneimittels aufgebracht werden, wodurch man einen dünneren Film für das Erhitzen erhält. Eine lineare Abnahme der Filmdicke steht im Zusammenhang mit einer linearen Abnahme der Verunreinigungen.
- Beispiel 3
- Zweites allgemeines Verfahren zum Verflüchtigen von Verbindungen Eine Flash-Anordnung aus einem äußeren Edelstahlzylinder und einer inneren Messingelektrode wurde in eine organische Lösung getaucht, die ein Arzneimittel enthielt, und schnell wieder daraus entfernt. Die Verdampfung von Lösungsmittelrückständen aus der Anordnung erfolgte durch Trocknen an der Luft. Dadurch blieb ein Film des Arzneimittels auf der Außenoberfläche des Edelstahlzylinders zurück. Die Anordnung wurde elektrisch mit einem Kondensatornetzwerk (z.B. 1,5 F) und einem mechanischen Relais unter Einsatz von Messinganschlüssen verbunden und in einem Glasmantel platziert. Eine Filteranordnung wurde zwischen dem Glasmantel und einem Vakuumsystem platziert. Der Fluss (~15 l/min) wurde durch den Glasmantel unter Einsatz des Vakuumsystems hervorgerufen. Das Erhitzen der Flash-Anordnung erfolgte etwa 0,25 s lang durch das momentane Einschalten des Relais zwischen der Flash-Anordnung und den Kondensatoren (mit Gleichstromquelle verbunden und mit einer Spannung von 20,5 V), um das aufgebrachte Arzneimittel zu verflüchtigen (ein Aerosol daraus zu bilden). Die Anordnung wurde abkühlen gelassen. Die Analyse des gebildeten Aerosols umfasste das Spülen des Filters mit 5 ml Acetonitril und das Injizieren einer Probe der organischen Lösung in eine HPLC. Sildenafilaerosol wurde unter Einsatz dieses Verfahrens in einer Reinheit von 98,9 % (0,075 mg) erhalten. Vardenafilaerosol wurde unter Einsatz dieses Verfahrens in einer Reinheit von 81,4 % (0,7 mg) erhalten.
Claims (11)
- Zusammensetzung zur Verabreichung eines Arzneimittels gegen erektile Dysfunktion, umfassend ein Kondensationsaerosol, a) gebildet durch das Verdampfen eines Arzneimittels gegen erektile Dysfunktion unter Bedingungen, die wirksam sind, um Dampf des Arzneimittels gegen erektile Dysfunktion zu produzieren, und Kondensieren des Dampfes des Arzneimittels gegen erektile Dysfunktion, um Kondensationsaerosolteilchen zu bilden, und b) worin diese Kondensationsaerosolteilchen durch weniger als 10% von Abbauprodukten des Arzneimittels gegen erektile Dysfunktion gekennzeichnet sind und c) das Aerosol einen gewichtsmittleren aerodynamischen Durchmesser (MMAD) von unter 5 μm aufweist.
- Zusammensetzung nach Anspruch 1, worin das Verdampfen die Erwärmung eines dünnen Films des Arzneimittels gegen erektile Dysfunktion auf einem festen Träger auf eine Temperatur umfasst, die ausreicht, um das Arzneimittel gegen erektile Dysfunktion vom dünnen Film zu verdampfen.
- Zusammensetzung zur Verabreichung von Sildenafil, Tadalafil oder Vardenafil, umfassend ein Kondensationsaerosol, a) gebildet durch das Verdampfen von Sildenafil, Tadalafil oder Vardenafil unter Bedingungen, die wirksam sind, um Dampf von Sildenafil, Tadalafil oder Vardenafil zu produzieren, und Kondensieren des Dampfes von Sildenafil, Tadalafil oder Vardenafil, um Kondensationsaerosolteilchen zu bilden, und b) worin diese Kondensationsaerosolteilchen durch weniger als 10% von Abbauprodukten der Arzneimittel Sildenafil, Tadalafil oder Vardenafil charakterisiert sind und c) das Aerosol einen MMAD von unter 5 μm aufweist.
- Zusammensetzung nach Anspruch 1, 2 oder 3, worin das Arzneimittel in Form einer freien Base vorliegt.
- Zusammensetzung nach Anspruch 3 oder 4, worin die Kondensationsaerosolteilchen zumindest 95 Gew.-% Sildenafil, Tadalafil oder Vardenafil umfassen.
- Verfahren zur Herstellung von Sildenafil, Tadalafil oder Vardenafil in Aerosolform, umfassend a) Verdampfen von Sildenafil, Tadalafil oder Vardenafil unter Bedingungen, die wirksam sind, um Dampf von Sildenafil, Tadalafil oder Vardenafil zu bilden, b) Ermöglichen des Abkühlens des Dampfes, wodurch ein Kondensationsaerosol gebildet wird, und c) Leiten von Luft durch den Dampf, um die Kondensationsaerosolteilchen von Sildenafil, Tadalafil oder Vardenafil zu transportieren, die weniger als 10 % von Sildenafil-, Tadalafil- oder Vardenafilabbauprodukten umfassen und einen MMAD von unter 5 μm aufweisen.
- Verfahren nach Anspruch 6, worin das Verdampfen das Erhitzen eines dünnen Films, der Sildenafil, Tadalafil oder Vardenafil umfasst und sich auf einem festen Träger befindet, auf eine Temperatur umfasst, die ausreicht, um Sildenafil, Tadalafil oder Vardenafil vom dünnen Film zu verdampfen.
- Verfahren nach Anspruch 6 oder 7, worin die Aerosolteilchen in einer Geschwindigkeit gebildet werden, die größer als 0,5 mg/s ist.
- Verfahren nach Anspruch 6, 7 oder 8, worin die Aerosolteilchen zumindest 70 Gew.-% Sildenafil, Tadalafil oder Vardenafil umfassen.
- Verfahren nach Anspruch 9, worin die Aerosolteilchen zumindest 95 Gew.-% Sildenafil, Tadalafil oder Vardenafil umfassen.
- Set zur Verabreichung eines wie in Anspruch 3 definierten Sildenafil-, Tadalafil- oder Vardenafilkondensationsaerosols, wobei das Set Folgendes umfasst: a) eine Zusammensetzung, umfassend Sildenafil, Tadalafil oder Vardenafil, und b) eine Vorrichtung, die aus der Zusammensetzung ein Sildenafil-, Tadalafil- oder Vardenafilkondensationsaerosol bildet, und worin die Vorrichtung (i) ein Element zur Erwärmung der Sildenafil-, Tadalafil- oder Vardenafilzusammensetzung zur Bildung eines Dampfes, (ii) ein Element, welches das Abkühlen des Dampfes ermöglicht, um ein Kondensationsaerosol zu bilden, und (iii) ein Element, das die Inhalation des Aerosols ermöglicht, umfasst.
